Antibody-drug conjugates and bispecific antibodies in the treatment of lymphomas
Lymphoma treatment has expanded substantially in recent years with antibody-drug conjugates and T-cell-engaging bispecific antibodies. These agents exploit surface antigens on malignant cells either to deliver a cytotoxic payload or to redirect T-cell activity and are progressively moving from later treatment lines into earlier therapeutic settings. This review summarizes the mechanism of action, pivotal clinical data, safety profile and current role of polatuzumab vedotin, brentuximab vedotin, loncastuximab tesirine and CD20×CD3 bispecific antibodies. Particular emphasis is placed on studies that established their approved indications and on practical differences relevant to treatment selection in routine clinical practice.
