Erdafitinib in treatment of advanced urothelial carcinoma

The current standard of care for advanced urothelial carcinoma in the first line is the combination of a conjugate with immunotherapy. In case of unavailability, platinum-based chemotherapy with maintenance avelumab if no progression, or pembrolizumab in case of progression on chemotherapy, are the options. Enfortumab vedotin is indicated following chemotherapy and immunotherapy failure. In patients with an identified gene alteration of the fibroblast growth factor receptor, the kinase inhibitor erdafitinib is an option. In this review, the mechanism of action, efficacy and safety in clinical trials, as well as management of erdafitinib adverse events are discussed.
